Effect of hypoglycemic sulphonylureas on vagal functions: ANIMALS AND METHODS


The experiments were carried out in 42 young mongrel dogs of both sexes weighing between 10 and 24 kg. The animals were anesthetized with 133 ^mol/kg pentobarbital (Phylaxia-Sanofi, Budapest, Hungary). The left vagal nerve was exposed and electrically stimulated with 1 ms long trains of square pulses of 2 V amplitude for 2 mins with increasing frequency (1, 2, 4, 8 and 16/s) by stimulator (Type ST-02, Experimetria, Budapest, Hungary).Mean arterial blood pressure was measured in the abdominal aorta through a cannula introduced through the femoral artery by Stoelting gauge (P23 Db). Heart rate was also detected. The changes of both parameters were registered on a multiscriptor (Medicor-CH-61, Medicor RT, Budapest, Hungary).
After the basal values were recorded, the alterations in blood pressure and heart rate were detected during the electrical stimulation of the vagal nerve 30 mins after intravenous administration of solvent.Changes in the above parameters are expressed as a percentage of the basal value.Electrical stimulation of the vagal nerve was repeated 30 mins after the intravenous pretreatment with the following sulphonylureas: carbutamide, tolbutamide (0.1, 1, 10, 100 and 1000 |j.mol/kg); chlorpropamide (0.1, 1, 10 and 100 |imol/kg); gliclazide, glimeripide (0.01, 0.1, 1, 10 and 100 |imol/kg); glipizide (0.001, 0.01, 0.1,1 and 10 |imol/kg); and glibenclamide (0.01, 0.1, 1, 10, 100 |imol/kg). Sulphonyl-ureas were dissolved in 10% ethanol and 10% polyethylene glycol in saline (pH 8). During the control investigation the animals were treated only with solvents.
